Born in Boston, Hillsmith studied began her art studies at the Boston Museum School of Fine Arts. She later studied in New York City at the Art Students League with John Sloan, Yasuo Kuniyoshi, Alexander Brook, William Zorach, and Vaclav Vytlacil. Hillsmith also spent several years working at Stanley William Hayter's New York City Atelier 17.

Hillsmith was both a respected artist and teacher. In 1945, she was invited by Josef Albers to teach at Black Mountain College for a season.
